摘要

Abstract

Aim:To investigate the effect of palbociclib on the metabolomics of cervical cancer Hela cells.Methods:Hela cells were treated with palbociclib at concentrations of 0.00,0.25,0.50,and 1.00 μmol/L for 24,48,and 72 hours,and cell proliferation was assessed using MTT assay.Cells treated with 0.00 and 0.50 μmol/L palbociclib for 72 hours were collected for metabolomic analysis via high-resolution liquid chromatography-mass spectrometry(HR-LCMS).The differenti-al metabolites between the 2 groups were compared and screened,and functional enrichment analysis of metabolic pathways was performed using the KEGG analysis.Results:Palbociclib significantly inhibited the proliferation of Hela cells,with the inhibitory effect enhancing in a dose-dependent manner(P<0.05).HR-LCMS identified 39 cationic differential metabolites and 24 anionic differential metabolites.Enrichment analysis indicated that the primary cationic metabolic pathway was the dopaminergic synapse pathway,while the primary anionic metabolic pathway was fatty acid biosynthesis.Conclusion:Palbo-ciclib could inhibit the proliferation of Hela cells,and the mechanism may involve the inhibition of fatty acid biosynthesis and disruption of the dopaminergic synapse pathway.
