Effect of Feiwei Granules on pulmonary fibrosis,autophagy,and epithelial mesenchymal transition in mice with pulmonary fibrosis

摘要

Abstract

Aim:To investigate the effect and potential mechanism of Feiwei Granules on pulmonary fibrosis in mice.Methods:Thirty-two mice were randomly divided into sham operation group,model group,positive control group,and Feiwei Granules group,with 8 mice in each group.A pulmonary fibrosis model was established by intratracheal injection of bleomy-cin in the latter 3 groups.From the 1st day after modeling,the sham operation group and model group received intratracheal injection of an equal volume of normal saline;the positive control group was given rapamycin(4 mg/kg)by gavage;the Fei-wei Granules group was given Feiwei Granules(13 mL/kg)by gavage.All interventions lasted for 2 weeks.Masson staining was used to observe collagen deposition;transmission electron microscopy was performed to count autophagosomes in lung tissue;ELISA was used to detect the levels of TGF-β1 and hydroxyproline in lung tissue;immunofluorescence staining was applied to measure the levels of epithelial mesenchymal transition markers(E-cadherin and α-SMA);Western blot was used to determine the levels of autophagy-related indicators(mTOR,Beclin-1,LC3 Ⅱ,and P62)in lung tissue.Results:Com-pared with the sham operation group,the model group showed an increased percentage of collagen deposition area in lung tissue,reduced autophagosomes,elevated levels of TGF-β1,hydroxyproline,α-SMA,mTOR,and P62,and decreased levels of E-cadherin,Beclin-1,and LC3 Ⅱ(all P<0.05).Compared with the model group,the positive control group and Feiwei Granules group exhibited a decreased percentage of collagen deposition area in lung tissue,increased autophagosomes,re-duced levels of TGF-β1,hydroxyproline,α-SMA,mTOR,and P62,and increased levels of E-cadherin,Beclin-1,and LC3 Ⅱ (all P<0.05).Conclusion:Feiwei Granules may ameliorate pulmonary fibrosis by synergistically regulating autophagy and epithelial mesenchymal transition.
