葛根素对肺腺癌A549细胞增殖和TOPK活性的抑制作用
Inhibitory effect of puerarin on proliferation and TOPK activity in lung adenocarcinoma A549 cells
摘要
Abstract
Aim:To investigate the effect of puerarin on the proliferation of lung adenocarcinoma A549 cells and its po-tential mechanism.Methods:The binding of puerarin to TOPK was assessed by microscale thermophoresis.The inhibitory effect of puerarin on TOPK activity was evaluated using in vitro kinase assay.A549 cells were treated with 0,25,50,100,and 200 μmol/L puerarin for 24 hours,the cell viability was measured by MTT assay,and the proliferative capacity was as-sessed via soft agar colony formation assay 14 days after treatment with 0,25,50,and 100 μmol/L puerarin.The phosphoryl-ation levels of TOPK,Histone H3,and ERK1/2 proteins were examined by Western blot.A549 cells were allocated into 2 groups and infected with lentiviral shTOPK or shMock,cells were treated with 100 μmol/L puerarin for 24 hours,and cell viability as well as the phosphorylation levels of TOPK,Histone H3,and ERK1/2 proteins were examined.Results:Puerarin directly bound to TOPK and inhibited its activity.Increasing puerarin concentrations reduced A549 cell viability,colony for-mation ability,and phosphorylation levels of TOPK,Histone H3,and ERK1/2(P＜0.05).Compared with the shMock group,cells in the shTOPK group exhibited further declines in cell viability and phosphorylation levels of TOPK,Histone H3,and ERK1/2(P＜0.05).Conclusion:Puerarin may inhibit A549 cell proliferation by directly targeting TOPK and sup-pressing its activity.关键词
