红松PkLEC1基因克隆及表达分析
Cloning and expression analysis of PkLEC1 gene in Korean pine
摘要
Abstract
The LEC1 gene is a key regulator in plant embryonic development.Cloned the Pinus koraiensis PkLEC1 gene,analyzed its protein structure and expression pattern,and laid the foundation for functional studies of this gene in somatic embryogenesis of P.koraiensis.The results showed that the cDNA of PkLEC1 was 540 bp in length,encoding 180 amino acids.Bioinformatics analysis indicated that PkLEC1 is an unstable hydrophilic protein without transmembrane domains or signal peptides,and subcellular localization predicts that it localizes to the nucleus.Sequence alignment and phylogenetic analysis revealed that PkLEC1 showed the closest homology to PsHAP3 of Pinus sylvestris and PcHAP3 of Pinus contorta.Gene expression analysis demonstrated that PkLEC1 gene was only highly expressed in callus with high embryonic potential and columnar embryonic-stage seeds.These findings suggest that PkLEC1 may contribute to maintaining embryogenic potential in P.koraiensis callus and regulating seed development during the columnar embryo period.关键词
