减数分裂相关基因在宫颈癌中应用价值的生信分析
Bioinformatics analysis of the value of meiosis-related genes in cervical cancer

Abstract
Objective:To explore the meiosis-related genes related to cervical cancer through bioinformatics methods and to analyze their clinical application value.Method:Based on the TCGA database,discover the differentially expressed genes between cervical cancer tis-sue and normal tissue.Use gene set enrichment analysis to screen out meiosis related genes.Based on the GEO database,screen the transcriptome sequencing data containing different dis-ease levels,different HPV infection states,and different radiotherapy and chemotherapy respon-ses,and perform secondary statistical analysis on them.Results:A total of 119 genes related to meiosis were screened,of which 14 were up-regulated in cervical cancer tissues and 5 were down-regulated.Among the differentially expressed meiosis-related genes,SYCE2,BLM,RBBP8,and RPA2 genes were finally found to be related to the prognosis of cervical cancer pa-tients.By calculating the Tau index,it was finally found that the three genes SYCP2,SYCE2,and SMC1B were specifically expressed in testicular/cervical cancer tissues.The area under the receiver operating characteristic(ROC)curve(AUC)for the diagnosis of CIN2+and CIN3+by SYCP2 was 0.846(95%CI:0.777~0.915),0.806(95%CI:0.731~0.881),respective-ly.The expression of SYCP2 in normal cervical tissues and low-grade lesions was higher than that in high-grade lesions and cervical cancer tissues.The expression levels of SYCP2 and SMC1B in HPV-positive cervical cancer were higher than that in HPV-negative patients,and the expression level of SYCE2 in HPV16-positive cervical cancer was higher than that in HPV-negative patients(all P<0.05).The expression levels of SYCP2 and SYCE2 in patients with complete response to radiotherapy and chemotherapy were lower than those with incomplete re-sponse(all P<0.05).Conclusion:Meiosis related genes SYCP2,SYCE2 and SMC1B,as testis/cervical cancer specific expression genes,are associated with HPV infection,and the expression level of SYCP2 is significantly positively correlated with the grade of cervical lesions,and the high expressions of SYCP2 and SYCE2 is associated with the resistance of cervical cancer to ra-diotherapy and chemotherapy.They are expected to be effective molecular targets for early screening and targeted therapy of cervical cancer.关键词
