Abstract

Isodon suzhouensis(Wangzaozi)is a medicinal plant belonging to the Lamiaceae family and is a geogra-phical indication product of Suzhou City.It has been traditionally used in folk medicine to treat various diseases.In this study,aiming to conduct genome sequencing of Isodon suzhouensis and explore genes related to its biosynthesis.A flow cytometry method to detect the genome size of Isodon suzhouensis was established by using Nicotiana benthamiana,Arabidopsis thaliana,and Salvia miltiorrhiza as reference plants.This study also compared the genome size of Isodon suzhouensis to other plants in the Labiaceae family to determine its phylogenetic relationship.The stu-dy suggests that Galbraith is a more suitable extraction solution for isolating nuclei from Isodon suzhouensis compared to OTTO extraction solution.The genome size of Isodon suzhouensis was found to be approximately 430 Mb,which is similar to the genome size of Rabdosia rubescens,indicating a close relationship between the two species.These fin-dings provide valuable insights for future genome sequencing of Isodon suzhouensis.
