Commentary on the first total synthesis of a complex sesterterpenoid calidoustene isolated from aspergillus calidoustus

Abstract
Sesterterpenoids represent a structurally complex and diverse class of natural products with a wide range of bioactivities.However,their limited natural availability has hindered in-depth studies.Total synthesis offers a viable and effective approach to access these structurally intricate molecules.This account describes the first total synthesis of a complex sesterterpenoid calidoustene isolated from the fungus Aspergillus calidoustus.The synthesis features an elegant retrosynthetic design and employs a modular synthetic strategy.A key step involves the construction of the densely functionalized C-ring via an intramolecular metal-hydride hydrogen atom transfer(MHAT)-initiated radical cyclization.This approach significantly streamlined the synthetic route.The work establishes a solid material foundation for further research into this class of sesterterpenoids.关键词
