犊牛轮状病毒TaqMan荧光定量PCR快速检测方法的建立
Establishment of a Rapid Detection Method for Calf Rotavirus Using TaqMan Fluorescence Quantitative PCR

Abstract
By analyzing the VP6 gene sequence characteristics of BRV in GenBank,this study successfully optimized specific primers and probes,established standard curves,and comprehensively validated the method's sensitivity,specificity,and reproducibility.The results showed excellent linear correlation between the Ct values of plasmid standards and their concentrations(R2=0.999 5),with a detection limit as low as 2.18×101 copies/μL.Additionally,the method only detected the specific amplification curve of BRV plasmid standards,with no fluorescence signal from negative controls,indicating good specificity.The coefficient of variation was less than 1%,suggesting excellent reproducibility.When applied to clinical samples,the method achieved a positive detection rate of 14.0%for porcine rotavirus infection,compared to 12.0%using conventional PCR methods.This method is suitable for clinical testing.关键词
