摘要

Abstract

Objective:To explore the intervention mechanism of Yijing Decoction on Diabetic Microvascular Abnormali-ties via the Angiopoietin(Ang)-Tunica intimal endothelial tyrosine kinase 2(Tie2)/Vascular endothelial growth factor(VEGF)Signaling Axis.Methods:Forty 12-week-old male SPF-grade rats(10 Wistar rats and 30 GK spontaneous dia-betic rats)were used.Wistar rats served as the normal group,while the 30 GK rats were randomly divided into the model group,western medicine group,and Yijing Decoction group,with 10 rats per group.The normal group and model group received PBS buffer via gavage.The western medicine group was administered calcium dobesilate tablets(156 mg/kg·d)diluted in PBS buffer via gavage.The Yijing Decoction group received Yijing Decoction water extract(10.71 g/kg·d of combined raw herbal dosage)via gavage once daily.All treatments lasted for 12 weeks.After treat-ment completion,all rats were euthanized,and eyeballs were enucleated to collect retinal tissues.RT-PCR was per-formed to detect mRNA expression of Ang1,Ang2,Tie1,Tie2,VEGF,Collagen Ⅳ(CⅣ),and Occludin.Western blot was used to analyze protein expression of Ang2,Tie2,VEGF,CⅣ,and Occludin.Results:In the model group,the mR-NA expression levels of Ang1,Tie2,and Occludin were significantly decreased compared to the normal group,while the mRNA expression levels of Ang2,Tie1,VEGF,and CⅣ were significantly increased,with statistically significant differ-ences(P<0.05).In both the Western medicine group and the traditional Chinese medicine(TCM)group,the mRNA expression levels of Ang1,Tie2,and Occludin were significantly upregulated compared to the model group,whereas the mRNA expression levels of Ang2,Tie1,VEGF,and CⅣ were significantly downregulated,with statistically significant differences(P<0.05).At the protein level,the model group exhibited significantly elevated Ang2,VEGF,and CⅣ ex-pression and reduced Tie2 and Occludin expression compared to the normal group(P<0.05).In contrast,both the Western medicine and TCM groups showed significantly decreased Ang2,VEGF,and CⅣ protein expression and in-creased Tie2 and Occludin protein expression compared to the model group,with statistically significant differences(P<0.05).Conclusion:Yijing Decoction alleviates early-stage diabetic microvascular damage and repairs inner blood-reti-nal barrier(iBRB)function by suppressing aberrant expression of Ang2,VEGF,and CⅣ while upregulating the expres-sion of Ang1,Tie2,and Occludin.
