五味麝香丸"卡嚓"方解热镇痛抗炎作用研究
Investigation on the Antipyretic,Analgesic and Anti-Inflammatory Efficacy of Wuwei Shexiang Pill"Ka Cha"Formula

Abstract
Objective:To investigate the main chemical constituents of Wuwei Shexiang pill"Ka Cha"formula and its antipyretic,analgesic,as well as anti-inflammatory effects.Methods:The chemical constituents of the Wuwei Shexiang pill"Ka Cha"formula were qualitatively detected using liquid chromatography-mass spectrometry(LC-MS).The acute toxicity test in mice was carried out using the maximum dosage(MAD)method to determine the pharmacological dosa-ges.Glacial acetic acid-induced writhing in mice was used to study its analgesic effect.Xylene-induced auricular swelling in mice was used to study the anti-inflammatory effect.The antipyretic effect was investigated in a mouse model of fe-ver caused by dry yeast.The levels of superoxide dismutase(SOD)and malondialdehyde(MDA)in the serum were de-tected,and the mRNA expression levels of inflammatory factors TNF-α,NF-kappa B,TLR4,COX2,and iNOS in the brain tissue were detected by qRT-PCR.Results:The Wuwei Shexiang pill"Ka Cha"formula contains 61 chemical components,including Quercetin,Epigallocatechins,and Asiatic acid.There were no deaths in the mice in the results of the maximum dosing experiments,no significant changes in the general behavioral examinations,and no damage in the histopathology of the liver and kidneys.In the analgesic experiment,the low,medium,and high doses of Wuwei Shex-iang pill"Ka Cha"formula significantly reduced the number of writhing mice caused by glacial acetic acid.In the anti-inflammatory experiment,the medium and high doses of Wuwei Shexiang pill"Ka Cha"formula significantly reduced the degree of auricular swelling in mice caused by xylene and significantly improved the pathological damage of ear tis-sue.In the antipyretic experiment,the doses of Wuwei Shexiang pill"Ka Cha"formula significantly reduced the fever caused by dry yeast in mice,increased the SOD content in serum to varying degrees,decreased the MDA content,and reduced damage to the hypothalamus tissue.The qRT-PCR results showed that Wuwei Shexiang pill"Ka Cha"formula could inhibit the increase in the mRNA expression levels of TNF-α,NF-kappa B,TLR4,COX2 and iNOS in the brain tissue of mice caused by dry yeast.Conclusion:Wuwei Shexiang pill"Ka Cha"formula has certain anti-inflammatory,analgesic and antipyretic effects,providing a theoretical basis for its clinical application and further research.关键词
