球姜酮通过上调Nrf2/HO-1信号通路缓解铁死亡保护小鼠溃疡性结肠炎机制研究
Study on the Mechanism of Alleviating Ferroptosis and Protecting Mice with Ulcerative Colitis by Upregulating the Nrf2/HO-1 Signaling Pathway with Curcumin

Abstract
Objective:Exploring the therapeutic effect and potential mechanism of galangamine on ulcerative colitis(UC)mice.Methods:Using dextran sulfate sodium salt(DSS)to induce a UC model,the therapeutic effect of curcu-min(20 mg/kg and 40 mg/kg)on UC was observed by oral gavage;Observe the structure of colon tissue through He-matoxylin Eosin(H&E)staining,and detect the levels of inflammatory factors in colon tissue through enzyme-linked immunosorbent assay(ELISA);Observe the levels of reactive oxygen species(ROS)in colon tissue through DHE stai-ning;By detecting the iron ion concentration,malondialdehyde(MDA)content,and glutathione(GSH)level in colon tissue,iron death related markers can be observed;Observation of mitochondria in mouse intestinal epithelial cells by transmission electron microscopy;Western blot was used to detect the expression of tight junction proteins(zonula oc-cludins 1(ZO-1)and occludin),as well as iron death related proteins glutathione peroxidase 4(GPX4),cysteine/gluta-mate reverse transporter(SLC7A11),nuclear factor-E2-related factor 2(Nrf2),and heme oxygenase-1(HO-1).Results:Globuline(20 mg/kg and 40 mg/kg)can significantly prolong colon length and reduce disease activity index(DAI)scores(P<0.01);Globuline(20 mg/kg and 40 mg/kg)can promote the expression of tight junction proteins(ZO-1 and Occludin)in colon tissue(P<0.01),maintaining the integrity of colonic villi,crypts,and goblet cells;Glob-uline(20 mg/kg and 40 mg/kg)can reduce inflammatory factors(tumor necrosis factor)in colon tissue-α(tumor nec-rosis factor-alpha,TNF-α)、Elevated levels of interleukin-1β and IL-6,ROS fluorescence intensity,iron ion concentra-tion,and MDA content,as well as elevated levels of IL-10 and GSH(P<0.01);The transmission electron microscopy results showed that curcumin(20 mg/kg and 40 mg/kg)significantly reduced mitochondrial damage.Globuline(20 mg/kg and 40 mg/kg)significantly promoted the expression of iron death related proteins GPX4 and SLC7A11(P<0.01);Globuline(20 mg/kg and 40 mg/kg)significantly activated the protein expression of Nrf2 and HO-1.Con-clusion:Zerumbone alleviates DSS-induced ulcerative colitis in mice by activating the Nrf2/HO-1 signaling pathway,thus inhibiting intestinal mucosal damage,inflammation,and iron death.关键词
