Abstract

In order to develop a highly effective diagnostic reagent for porcine epidemic diarrhoea virus(PEDV),a porcine epidemic diarrhoea virus blocking ELISA antibody detection kit was developed by preparing pre-coated microplates and enzyme labelled antibodies,and specific tests,neutralising activity tests and clinical diagnostic tests were carried out to verify the performance of the kits.The results showed that the monoclonal antibody prepared by the kit had good specificity and only reacted positively with PEDV-related strains,and had no cross-reactivity with other strains and cells.Each monoclonal antibody had high neutralising activity against different PEDV strains,such as PEDV-Mabs-2D5 against PEDV-KB2013-4 strains with a neutralising titre of 1∶2 048,the minimum detection amount of the kit was 1∶256,and the specificity coincidence rate was 100%,and the clinical test results were consistent with expectations.In summary,the ELISA antibody detection kit for swine epidemic diarrhoea virus blockade prepared in this experiment has strong specificity and high sensitivity,which is of great significance for judging the PEDV infection situation in pig farms and evaluating the immunisation status of swine fever vaccine,and provides an effective detection method for PEDV prevention and control.
