Isolation,and Identification of Pasteurella multocida from Clinical Swine Samples in Different Regions and Screening of Highly Virulent Strains

Abstract
To investigate the prevalence and virulence of Pasteurella multocida(Pm)in Chinese swine farms,suspected nasal swabs and lung tissues from diseased pigs in farms across Henan,Zhejiang,and Jiangsu provinces were collected for pathogen isolation and identification.Gram staining,biochemical identification,PCR serotyping,antimicrobial susceptibility testing,mouse pathogenicity tests,and piglet challenge trials were performed.A total of six Pm strains were isolated,including four serotype A and two serotype D strains.All isolates were sensitive to polymyxin B,doxycycline,enrofloxacin,ciprofloxacin,gentamicin,and ceftiofur,while resistant to bacitracin,clindamycin,and lincomycin.Through mouse pathogenicity assays,a highly virulent strain P1971-A was identified.Piglet challenge experiments showed that a dose of 1×10⁷ CFU/head caused illness in 40%of piglets,while 1×10⁹ CFU/head led to 100%morbidity and 60%mortality.Infected piglets displayed typical clinical signs including lethargy,reduced appetite,recumbency,and dyspnea.Necropsy of deceased piglets revealed tracheal white froth and pulmonary emphysema.These findings demonstrate that four serotype A and two serotype D strains of Pm were successfully isolated,providing valuable insights into the prevailing serotypes and pathogenicity of Pm strains in China,and offering reference data for vaccine development.关键词
