Identification,Bioinformatics,and Functional Analysis of Porcine Gasdermin B Isoforms

Abstract
Gasdermin B(GSDMB),a member of the gasdermin(GSDM)family,plays a crucial regulatory role in infectious diseases,cancer,and inflammatory disorders.To elucidate the tissue distribution,isoform characteristics,and functional activity of porcine GSDMB(pGSDMB),this study employed real-time fluorescent quantitative polymerase chain reaction(qPCR)to determine its transcription levels of pGSDMB in various cell lines and tissues.PCR was used to clone and identify its isoforms,followed by bioinformatics-based protein structure prediction and analysis.The pyroptotic activity of the N-terminal domain was assessed using lactate dehydrogenase(LDH)release assay and propidium iodide(PI)staining.qPCR results revealed that pGSDMB was highly expressed in porcine ileum epithelial cell line(IPI-2I cells)and intestinal tissues.Two isoforms of pGSDMB,namely pGSDMB2 and pGSDMB3,were identified for the first time.Structural analysis showed that pGSDMB2 lacks a 21-amino acid structural region(comprising a β-sheet and part of an unstructured loop)compared to pGSDMB3.Functional assays confirmed that the N-terminal domain of pGSDMB(amino acids 1-245)exhibited significant pyroptotic activity compared to the empty vector control(P<0.000 1),whereas the truncated N-terminal fragment(1-222 amino acids)lacking 21 residues lost this function.In conclusion,this study characterized the transcriptional profile of pGSDMB across different cells and tissues,revealed sequence differences and structural features of its isoforms,and identified the functional pyroptotic region within the N-terminal domain(1-245 aa).These findings provide important insights into the biological function of pGSDMB and its potential role in disease pathogenesis.关键词
