小麦隐性抗白粉病基因pmCH1853的遗传分析与分子定位
Genetic analysis and molecular mapping of a recessive powdery mildew resistance gene pmCH1853 in wheat

Abstract
To screen for new powdery mildew resistance genes in wheat,phenotypic identification and genetic analysis were conducted on the genetic population derived from the resistant parent CH1853 and susceptible parent Taichang 29.The bulked segregant analysis and 90K single nucleotide polymor-phism(SNP)chip technology were employed for genetic mapping of the powdery mildew resistance gene in the resistant parent CH1853.The results showed that the F1 plants derived from the resistant parent CH1853 and susceptible parent Taichang29 were susceptible to Blumeria graminis f.sp.tritici isolate E09,and their F2 individuals showed segregation of resistance.The ratio of resistant individuals to susceptible individuals roughly followed a 1∶3 segregation ratio in F2 population,indicating that the powdery mildew resistance was controlled by a recessive gene,temporarily named as pmCH1853.A total of 89 SNPs exhibited polymorphism between the resistance bulk and susceptible bulk,of which 44 SNPs were located on chromosome 2D,and 42 SNPs were distributed within the 635.6-650.3 Mb genomic interval.Five simple sequence repeat markers linked to resistance were obtained within this region.Eventually,pmCH1853 was mapped between markers Sxau2DL-115 and Sxau2DL-147,with genetic distances of 0.8 cM and 3.2 cM,respectively.The physical location was narrowed down to the 635.6-639.9 Mb interval.This gene can be used as a novel genetic resource for powdery mildew resis-tance in wheat breeding programs.关键词
