Abstract

To obtain biocontrol bacteria for the control of banana Fusarium wilt disease,antagonistic bacteria with good inhibitory effects on Fusarium oxysporum f.sp.cubense race 4(Foc4)were isolated from the healthy banana rhizosphere soil using plate confrontational culture method.The antagonistic strain was identified by a combination of morphological observation,physiological and biochemical characteristics,and phylogenetic analysis of multiple genes including 16S rDNA,TrpB,RpoB,and GyrB.The anti-fungal spectrum,biological control and growth-promoting efficiency of the antagonistic strain were also investigated.The results showed that a total of 102 bacterial strains were isolated from the rhizosphere soil of banana plants.The strain GD3-16 had the highest antagonistic activity against Foc4 with an inhibition rate of 62.8%,which was further identified to be Kitasatospora recifensis with colony morphology,physiological and biochemical characteristics,and phylogenetic analysis of mul-tiple genes.The strain GD3-16 altered Foc4 hyphal morphology by causing malformation and enlarge-ment.The strain GD3-16 also inhibited spore germination of Foc4 significantly with an inhibition rate of 43.9%.Volatile organic compounds(VOCs)from the strain GD3-16 significantly inhibited fungal mycelial growth with an inhibition rate of up to 91.6%using the dual plate assay.Thirteen VOCs were identified through gas chromatography-mass spectrometry.The strain GD3-16 exhibited broad-spectrum antagonistic activity to ten phytopathogenic fungi tested.Furthermore,K.recifensis GD3-16 had significant control effects on banana Fusarium wilt with a biocontrol efficiency of 59.6%.The strain GD3-16 effectively promoted banana growth,which possessed abilities to degrade lignin,secrete proteases and produce siderophores.In conclusion,K.recifensis GD3-16 has the potential as the ideal biocontrol agent in controlling banana Fusarium wilt with high efficiency and plant growth-promoting properties.
