Abstract

Objective:To explore the effects of different treatment conditions on the loss and quality of polysaccharide in the pro-cessing of Polygonati Rhizoma(Huangjing).Methods:Natural drying,hot air drying,vacuum freeze-drying,rapid"nine"process,beater steaming,beater filtration steaming,whole root steaming and direct boiling were used to treat Huangjing.The polysaccharide content of Huangjing was determined by anthrone-sulfuric acid method,and Huangjing quality was analyzed by microorganism and heavy metal detection.Results:The polysaccharide content of fresh Huangjing was 11.5%.After natural drying,hot air drying,vacuum freeze-drying,rapid drying,beater filtration steaming,beater steaming,whole root steaming and direct boiling,the polysaccharide content of Huangjing was 10.23%,9.98%,12.08%,33.53%,28.9%,16.6%,28.7%and 21.8%,respectively.The microbial test showed that the total number of colonies,Escherichia coli and mold in all samples were<10 CFU·g-1,and no pathogenic bacteria were detected.Lead and cadmium were not detected(limit of quantification 0.04 mg·kg-1).Conclusion:The rapid"nine"process has the highest polysaccharide content retention rate,which is significantly better than the traditional method.Controlling drying conditions(mode,temperature,and time)can effectively reduce the loss of polysaccharide during the processing of Huangjing.
