Abstract

Objective:To investigate the regulatory mechanism of Wenhe Decoction on ferroptosis in hot water bath-induced febrile seizure(FS)model rats through SOD/Nrf2/HO-1 signaling pathway,thereby restoring Glu/GABA balance.Methods:Fifty SD rats were randomly divided into blank group,model group,and low-,medium-,high-dose Wenhe Decoction groups(10 rats each).After successful modeling,the three Wenhe Decoction groups received 4.05 g/kg,8.1 g/kg,and 16.2 g/kg(1∶2∶4 ratio)respectively via gavage(10 mL/kg);blank and model groups received equal-volume distilled water,once daily for21 days.Except blank group,rats were placed in(45±0.5)℃water bath until seizure onset,then immediately removed and dried;control group was placed in(37±0.5)℃bath for 5 minutes.Seizure latency,duration,and behavioral symptoms were recorded.Hippocampal pathology was observed via HE and Nissl staining.ROS levels were detected by DHE fluorescence probe.Serum iron,ATP,Glu,GABA,SOD,Nrf2,and HO-1 were measured by ELISA.Results:Compared with blank group,FS model rats showed shortened seizure latency,prolonged duration,increased seizure grade/incidence(P<0.05,P<0.01);hippocampal neurons exhibited loss,reduced count,irregular morphology,and disarrangement;serum iron,ROS,Glu,SOD,Nrf2,and HO-1 significantly increased(P<0.05,P<0.01),while ATP and GABA decreased(P<0.01).Compared with model group,all Wenhe Decoction doses improved behavioral symptoms,hippocampal neuron arrangement,and reduced pathological damage;serum iron,ROS,Glu,SOD,Nrf2,and HO-1 decreased(P<0.05,P<0.01),while ATP and GABA increased(P<0.01).Conclusion:Wenhe Decoction may activate SOD/Nrf2/HO-1 pathway to inhibit ferroptosis,restore Glu/GABA balance,reduce neuronal damage and FS recurrence.
