摘要

Abstract

Objective:To determine the content of main active components in serum containing Astragali Radix and Lophatheri Herba Couplet Medicinals at different time points by targeted metabolomics,so as to provide reference for subsequent in vitro experiments using serum prepared at the most appropriate time point.Methods:The Astragali Radix and Lophatheri Herba couplet medicinals decoction was administered to rats by gavage twice daily.On the 3rd and 7th days,sterile blood samples were collected from the abdominal aorta at 0.5 h,1 h,1.5 h,and 2 h after the last gavage to prepare drug-containing serum.Targeted metabolomics(LC-MS)was used to construct total ion current chromatograms(TIC)of drug-containing serum at different blood collection times.Based on the peak areas of characteristic peaks in TIC,the contents of astragaloside IV(active component of Astragali Radix)and ferulic acid(active component of Lophatheri Herba)in the drug-containing serum were calculated,and the data were analyzed to draw conclusions.Results:The content determination of astragaloside IV showed that the highest content was observed at 2 h after administration on the 7th day(S2),with significantly higher levels on the 7th day than on the 3rd day.The content determination of ferulic acid showed that the highest contents were observed at 0.5 h after administration on the 3rd day(T0.5)and 0.5 h after administration on the 7th day(S0.5),with significantly higher levels on the 7th day than on the 3rd day.Conclusion:The optimal blood collection time for preparing drug-containing serum of Astragali Radix and Lophatheri Herba couplet medicinals is 2 h after the last gavage on the 7th day of administration,which may provide reference for subsequent in vitro experiments.
