Abstract

Objective:To optimize the extraction process of polysaccharides from Fritillaria taipaiensis using a compound enzyme method and to explore its antioxidant activity.Methods:Based on preliminary experiments with a fixed solid-liquid ratio of 1∶15 g/mL,the extraction process was optimized using response surface methodology.The independent variables were the amount of compound enzyme,enzymatic hydrolysis time,enzymatic hydrolysis temperature,and solution pH,while the dependent variable was the polysaccharide extraction rate.The antioxidant activity of the extracted polysaccharides was evaluated by measuring their DPPH free radical scavenging ability and Fe3+reduction capacity.Results:The optimal extraction conditions were as follows:compound enzyme amount 2.3%,enzymatic hydrolysis time 63 minutes,pH 5.2,and enzymatic hydrolysis temperature 43℃.Under these conditions,the actual extraction rate of polysaccharides was 10.034 9%,which was very close to the predicted value of 10.050 1%,with a difference of only 0.015 2%,indicating that the model was reliable and could be used for predicting polysaccharide extraction.Antioxidant experiments showed that at a high concentration of 3.0 mg/mL,Fritillaria taipaiensis polysaccharides exhibited DPPH radical scavenging and Fe3+reducing abilities similar to those of the positive control(vitamin C)at the same concentration,with no significant differences.Conclusion:Fritillaria taipaiensis polysaccharides possess significant antioxidant activity,providing experimental evidence for the development and application of this medicinal herb.This study also offers new insights and methods for the application of polysaccharides in the fields of health food and medicine.
