RRM2过表达对苯肾上腺素诱导的AC16心肌细胞肥大及凋亡的影响
Effect of overexpression of RRM2 on phenylephrine-induced hypertrophy and apoptosis in AC16 cells

Abstract
Objective To explore the role of ribonucleotide reductase M2 subunit(RRM2)in phenylephrine(PE)-induced myocardial hypertrophy and apoptosis.Methods AC 16 cells were treated with 10,50 and 100 μM PE to screen the optimal con-centration of PE for inducing the cardiomyocyte hypertrophy model,and the expression of RRM2 was analyzed.The AC16 cardio-myocytes were divided into the following groups:lentivirus transfection negative control group(LV-NC),lentivirus transfection negative control+phenylephrine(PE)treatment(LV-NC+PE)group,and lentivirus overexpression of RRM2+phenylephrine treatment(LV-RRM2+PE)group.The western blot method and reverse transcription-quantitative polymerase chain reaction(RT-qPCR)method were used respectively to detect RRM2,myocardial hypertrophy markers(ANP,BNP,and β-MHC),and apoptosis-related markers(Bax and Bc12)in each group.Additionally,the cross-sectional area of cardiomyocytes was assessed using TRITC-labeled phalloidin in an immunofluorescence assay.Changes in the apoptosis rates of the cells were detected using flow cytometry.Results Treatment of AC16 cells with 50 μM PE for 48 hours could construct an ideal myocardial cell hypertro-phy model.The RNA and protein levels of RRM2 were significantly downregulated in the AC16 cardiomyocyte hypertrophy model(P<0.05 or P<0.01).Compared with that in the LV-NC+PE group,the cell area of AC16 in the LV-RRM2+PE group was significantly reduced(P<0.05).Overexpression of RRM2 could inhibit the mRNA and protein levels of cardiac hypertrophy markers ANP,BNP,and β-MHC(P<0.05),and reduce the proportion of PE induced cell apoptosis(P<0.01).Conclusion Overexpression of RRM2 can alleviate PE-induced hypertrophy and apoptosis in AC16 cells.关键词
