Study on Seed Morphological Structure and Germination Characteristics of Rhodiola crenulata

Abstract
Objective:To investigate the morphological structure and germination characteristics of Rhodiola crenulata(Hook.f.&Thomson)H.Ohba seeds,an endangered medicinal plant,and to provide a theoretical basis for its artificial propagation and resource conservation.Methods:The morphological characteristics of seeds were observed using a stereomicroscope,the anatomical structures of seeds were examined by means of paraffin sectioning combined with safranin-fast green staining,and the 1000-seed weight and water content of seeds were determined.The effects of temperature,light conditions,warm water soaking,and plant growth regulators on seed germination were systematically studied.Results:Rhodiola crenulata seeds are narrowly ovoid,winged,and exhibit a dicotyledon-ous exalbuminous structure.The thousand-seed weight was 0.110 g(moisture content 7.3%).The optimal germination temperature was 20℃(germination rate 94.7%,germination energy 79.4%).The seeds were categorized as positively photoblastic,with light significantly enhancing both germination rate and energy.Soaking in 50℃ water for 2 h significantly increased germination rate(98.8%)and energy(86.4%).Low concentrations of 6-BA(10-50 mg/L)significantly promoted germination(germination rate>97%,germination energy>81%).Conclusion:The seed germination of R.crenulata is significantly influenced by factors such as temperature,light,and hormones.The optimal germination temperature,light requirements,duration of warm water soaking,and hor-mone treatment conditions were preliminarily determined,providing key parameters for optimizing its artificial propagation techniques.关键词
