阳虚大鼠抑郁易感性分析及对前额叶乳酸化修饰的影响
Analysis of Depressive Susceptibility in Yang Deficient Rats and Effects on Prefrontal Lactylation

Abstract
Objective:To establish a rat model of Yang deficiency depression(YDD)by the bitter cold method combined with CUMS,to dynamically evaluate the susceptibility relationship of yang deficiency and depression,and to explore the common potential mechanism of the two based on lactate and lactylation.Methods:Yang deficiency(YD)rat model was firstly constructed by bitter cold method(Huangbai 3.24g/kg solution oral gavage+4℃ cold environment for 1 h/d)for 2 weeks,followed by superimposing 4 weeks of CUMS to construct the YDD rat model.Behavioral tests such as yang deficiency evaluation,sugar water preference,forced swimming,open field,and three-box so-cial text were performed at weeks 2,4,and 6,and prefrontal lactate,SIRT1,and lactylation levels were measured in rats at week 6.Re-sults:Compared with the blank group,the bitter cold method successfully established a rat YD model;the rats in the depressed group showed signs of Yang deficiency in the 6th week(P<0.01);the YDD rats showed a decrease in sugar water preference in the 4th week(P<0.05),and the depressed rats showed a decrease in sugar water preference in the 6th week(P<0.05),the depressed rats showed a de-crease in sugar water preference at week 6(P<0.05);YDD rats showed a significant decrease in total distance,number of grid crossings,and number of support stands at week 4(P<0.01),compared to the depression group(P<0.05);The open field immobility time of YDD rats significantly increased at week 4(P<0.05),while the depression group showed a significant increase in immobility time at week 6(P<0.01);At weeks 4 and 6,the number of forced swim fights significantly decreased in YDD(P<0.001)and depression group(P<0.01).The social time of depression group and YDD rats significantly decreased at week 6(P<0.05).The levels of lactate,SIRT1 and lactyla-tion in the prefrontal cortex of YDD rats were significantly decreased(P<0.05).Conclusions:The rats in the Yang deficiency and depres-sion groups showed similar phenotypes."Yang deficiency"induced or aggravated depression-like behaviors in rats,and reduced lactate/SIRT1-mediated modification of prefrontal lactylation may be a potential mechanism common to both Yang deficiency and depression.关键词
