石氏逐痰通络汤通过调控"骨-筋-肉"轴延缓自发膝骨关节炎豚鼠膝关节退变的病理机制研究
Study on Pathological Mechanism of Shi's Zhutan Tongluo Decoction in Delaying Spontaneous KOA Guinea Pigs Knee Joint Degeneration by Regulating The"Bone-Tendon-Muscle"Axis

Abstract
Objective:To investigate the effects of Shi's Zhutan Tongluo Decoction on cartilage degeneration,synovial inflammation and muscle degeneration in guinea pigs with spontaneous knee osteoarthritis(KOA)model,so as to clarify the mechanism of the pre-scription in protecting the knee joint of KOA guinea pigs.Methods:Strain 13 guinea pigs were selected as the blank group;Dunkin Hartley guinea pigs were selected as the spontaneous KOA model.They were randomly divided into the model group and Zhutan Tongluo Decoction group after 14 months of modeling.Guinea pigs were killed after 8 weeks of continuous administration,and knee cartilage,synovium and quadriceps tissues were obtained.Guinea pig knee cartilage,synovium and quadriceps were stained with he-matoxylin-eosin(HE),and knee cartilage was stained with toluidine blue.Mankin score,Krenn score and muscle tissue biopsy score were used to evaluate cartilage degeneration,synovial inflammation and quadriceps degeneration in guinea pigs.Results:The Mankin score of cartilage in the Zhutan Tongluo Decoction group was significantly lower than that in the model group(P＜0.05).Compared with the model group,it was observed that the articular surface of cartilage in the Zhutan Tongluo Decoction group was more complete,subchondral bone sclerosis was improved,and multiple tide lines were clearer;the Krenn score of synovium in the Zhutan Tongluo De-coction group was significantly lower than that in the model group(P＜0.05).Compared with the model group,it was observed that the synovial hyperplasia of guinea pigs in the Zhutan Tongluo Decoction group was improved,inflammatory cells were reduced,and vascular hyperplasia was relieved;the biopsy score of quadriceps muscle tissue in the Zhutan Tongluo Decoction group was significantly lower than that in the model group(P＜0.0001).Compared with the model group,it was observed that the muscle fibers of quadriceps muscle in the Zhutan Tongluo Decoction group were larger,the proportion of central nuclei was reduced,and interstitial fibrosis was re-lieved.Conclusion:Zhutan Tongluo Decoction can significantly improve the pathological degeneration of various tissues in the knee joint of spontaneous KOA guinea pigs,delay cartilage degeneration,reduce synovial inflammation,and improve muscle atrophy.关键词
