摘要

Abstract

In order to predict and analyze scenarios where ISCR elements provide promoters for downstream resistance genes,sequence characteristics of identified ISCR family elements and their captured resistance genes were examined.Three classical IS91 family members(IS801,IS1294,IS91)and 17 publicly availa-ble ISCR sequences were included.Phylogenetic analysis of I SCR transposase amino acid sequences was performed using MEGA X,and variations within conserved motifs were aligned.Conserved nucleotide fea-tures of oriIS and terIS termini were compiled from literature,including characteristic base-pair patterns.Complete reference I SCR sequences were retrieved from the NCBI GenBank database,preliminarily annota-ted via the RAST platform,and resistance genes were verified using the Resfinder database.Intergenic re-gions between ISCR elements and downstream resistance genes were analyzed with CLC Genomics Work-bench 8.Promoter prediction was conducted using BPROM for 200 bp sequence upstream of each resistance gene.Cluster analysis demonstrated that IS28 and IS20 exhibit the closest evolutionary proximity to IS91 family members.While oriIS termini were highly conserved,terIS termini-excluding internal reverse-complementary sequences-lacked significant nucleotide conservation.ISCR elements facilitated the capture of at least 20 resistance genes,with eight cases suggesting that ISCR right termini provide functional pro-moters.These results indicate that ISCR elements not only mobilize resistance genes but may also drive their expression through promoter provision.This functional distinction from IS26,coupled with its status as the second most active insertion sequence family(after IS26)in resistance gene transfer,underscores its critical significance.
