摘要

Abstract

To understand the genetic variation and evolution of porcine reproductive and respiratory syn-drome virus type 1(PRRSV 1)in China,by amplifying and sequencing,we acquired the full-length genome of a strong positive sample of PRRSV 1 from a large-scale pig farm in Guangdong province,and performed homology alignment and genetic evolution analysis compared to referenced strains.The results showed that the full-length genome of GD2023 was 14 979 bp(without poly A),with 86.7%and 60.6%nucleotide ho-mology with PRRSV 1 and PRRSV 2 classic strains LV and VR2332,respectively.The phylogenetic tree based on the complete genome showed that GD2023 was in the same branch with PRRSV 1 strains,sugges-ting that GD2023 belonged to PRRSV 1.ORF5 genetic evolution tree analysis showed that GD2023 was in the same branch with variant strain Lena,which was the closest to SL-01.Amino acid sequence comparison analysis of nsp2 showed that nsp2 had 43 amino acid deletions(aa289,aa320-aa360,and aa420),with a de-letion pattern 1+41+1.Furthmore,large difference was identified through analyzing the signal peptide,the potential glycosylation site,and the neutralization epitope of the GP5 protein.No recombination event was determined.Our results demonstrated that the mutation and evolution of PRRSV 1 is ongoing in the field,and much attention and monitoring need to be strengthened.
