Abstract

Primers and probes were designed based on the specific conserved sequences of the M gene of por-cine epidemic diarrhea virus(PEDV)and the S gene of porcine infectious gastroenteritis virus(TGEV),respectively,to establish a duplex fluorescence recombinase-mediated isothermal amplification(RAA)method for detecting PEDV and TGEV.This method can complete the detection at a constant temperature of 41 ℃ for 20 minutes,without cross reactivity with porcine coronavirus,porcine rotavirus,porcine circo-virus type 2,classical swine fever virus,and porcine pseudorabies virus.The minimum detection limits for PEDV and TGEV plasmid standards are 101 copies/μL and 103 copies/μL,respectively.The RT-qPCR de-tection method and the established fluorescent RAA detection method were used to detect 50 clinical sam-ples,and the positive coincidence rate of the two was 93.10%.The results indicated that the established duplex fluorescence RAA detection method for PEDV and TGEV has the potential to become an efficient diagnostic tool for veterinary clinical testing.
