NF-κB/NLRP3信号通路对丙泊酚致幼龄大鼠海马损伤的调节作用
Regulation Roles of the NF-κB/NLRP3 Pathway in Propofol-induced Hippocampal Injury in Young Rats

Abstract
This study aimed to elucidate the pivotal role of the NF-κB/NLRP3 signaling pathway in propofol-induced neurotoxicity during development.Seven-day-old SD rats were allocated into four groups:control group,propofol group(intraperitoneal injection five times,with an initial dose of 40 mg/kg fol-lowed by injections every hour),Bay11-7082 group(NF-κB inhibitor,injection at 5 mg/kg),and Bay11-7082+propofol group(Bay11-7082 injected 30 minutes prior to propofol anesthesia).Brain tissues were subjected to HE staining,and the expression levels of proteins associated with the NF-κB and classical py-roptosis pathways in the hippocampus were evaluated using Western blot analysis.The concentrations of cytokines IL-1β,IL-6,IL-10,and IL-18 were quantified via ELISA.The results of HE staining indicated that following propofol administration,significant histological damage occurred in the hippocampus of young rats,accompanied by the activation of the NF-κB/NLRP3 signaling pathway.Pretreatment with Bay11-7082 effectively mitigated the histological damage induced by propofol in the hippocampus,inhibited the expression of p-p65 protein,downregulated levels of pyroptosis-related proteins(NLRP3,caspase-1 p20,and cleaved GSDMD),reduced pro-inflammatory cytokine levels,and upregulated anti-inflammatory cytokine IL-10.Activation of the NF-κB/NLRP3 signaling pathway is implicated in propofol-induced py-roptosis and neuroinflammation within the hippocampus of young rats;furthermore,Bay11-7082 demon-strates efficacy in alleviating hippocampal injury caused by propofol.关键词
