Expression and regulation of key genes during early growth and development of Eocanthecona furcellata(Wolff)

Abstract
[Objective]During mass rearing of the Eocanthecona furcellata(Wolff),partial egg masses often failed to be hatched and early-stage nymphs(1st and 2nd instars)exhibited high mortality.This study aimed to identify key coding genes involved in the early growth and development of Eocanthecona furcellata(Wolff),and,to preliminarily construct a putative gene interaction regulatory network for early development based on expression profiles,so as to provide a theo-retical foundation for elucidating the molecular regulatory mechanisms of early development of Eocanthecona furcellata(Wolff)and its closely related insect species,and for optimizing large-scale rearing practices.[Method]Transcriptome se-quencing was performed to quantify gene expression in eggs,1st instar nymphs and 2nd instar nymphs of Eocanthecona furcellata(Wolff).Differentially expressed genes(DEGs)were identified between egg and 1st instar nymphs(eggs vs 1st instar nymphs),and between 1st instar nymphs and 2nd instar nymphs(1st instar nymphs vs 2nd instar nymphs).Based on the expression of DEGs and functional annotations of Eocanthecona furcellata(Wolff)genome,key coding genes in-volved in early developmental processes were analyzed.Furthermore,10 key genes and transcription factors were ran-domly selected for validation using quantitative real-time PCR(qRT-PCR).[Result]In the group of eggs vs 1st instar nymphs,2681 DEGs were identified between eggs and 1st instar nymphs,while 1014 DEGs were identified between 1st in-star nymphs and 2nd instar nymphs;in the group of 1st instar nymphs vs 2nd instar nymphs,there was 2060 DEGs identi-fied between eggs and 1st instar nymphs and 393 DEGs were identified between 1st instar nymphs and 2nd instar nymphs;621 DEGs were shared by both groups.GO functional annotation and KEGG signal pathway enrichment analysis of DEGs identified 94 DEGs involved in the early growth and development of Eocanthecona furcellata(Wolff)and 5 transcription factors(AP-2-beta,HNF-4,sem-2,collier,and LBX1).Further analysis suggested that sexual development and wing development may start at the egg stage,while high expression was observed in DEGs associated with predatory behavior and digestive adaptation of 1st instar nymphs.And qRT-PCR results of selected genes were consistent with transcriptome analysis,confirming the reliability of transcriptomic analysis for detecting gene expression.[Conclusion]In this study,94 key coding genes associated with growth and development are identified,and 5 of which encoding transcription factor genes may synthetically regulate the early development,including digestion,saliva/venom production as well as olfaction of the Eocanthecona furcellata(Wolff).关键词
