Abstract

During the operation of an anaerobic ammonium oxidation(Anammox)reactor,dissolved oxygen(DO)inevitably enters the reactor,leading to the presence of nitrifying bacteria that compete with AnAOB for substrates,significantly hindering the enrichment of AnAOB.In this study,biofilm from a municipal wastewater treatment plant was used as inoculating sludge to start an Anammox reactor under micro-aerobic conditions,aiming to investigate the enrichment characteristics of AnAOB.The results showed that under a DO concentration of(1.0±0.2)mg·L-1,the apparent activity of AnAOB continuously increased with the rise of the nitrogen loading rate(NLR),reaching(682.42±43.88)mg·(L·d)-1,while the apparent activities of AOB and NOB initially increased and then decreased,eventually stabilizing.This indicates that AnAOB can be successfully enriched under micro-aerobic conditions.High-throughput sequencing revealed that while the reactor successfully enriched AnAOB-Ca.Brocadia(27.18%),it also enriched AOB-Nitrosomonas(0.38%)and NOB-Nitrospira(0.45%),demonstrating the inevitable presence of nitrifying bacteria in the system.Simulation results indicated that with the addition of exogenous NO2--N,even at a DO concentration of 5.0 mg·L-1,nitrifying bacteria in the biofilm surface layer could effectively prevent DO diffusion into the AnAOB layer,ensuring the successful proliferation of AnAOB.However,in the absence of exogenous NO2--N,when the DO concentration reached ≥ 3.0 mg·L-1,the enrichment of AnAOB biofilm was significantly restricted by DO,and nitrifying bacteria gradually dominated,severely inhibiting AnAOB proliferation.These findings provide valuable insights for the practical application of AnAOB technology in engineering projects.
