Different environmental factors on spermatozoa motility of Leuciscus waleckii from Dali Lake

Abstract
Leuciscus waleckii is one of the only two kinds of economic fish from Dali Lake,a high saline-alkali lake in Inner Mongolia.In recent years,reduced inflow into Dali Lake and drought conditions have led to a progressive increase in the alkalinity of Dali Lake.This environmental shift has been accompanied by a decline in the wild spawning population of L.waleckii.To understand the influence of environmental factors on the sperm motility of L.waleckii in Dali Lake,sperm activation rate,fast movement time and life time were used as evaluation indexes to screen out the optimal sperm activator(water of Gongger River,water of Dali Lake,different pH solutions,different concentrations of NaCl,KCl,MgCl2,NaHCO3 and glucose solutions)to improve the fertilization rate of artificial reproduction.The results showed that the average life time of sperm was(843.33±100.49)s,and the fast movement time was(248.33±22.10)s after lake water sperm activation,and the life time was nearly 8 times that of river water activation.With the increase of pH,the fast movement time and life time of the sperm increased first and then decreased.When the pH was 8,the fast movement time(1 254.00±166.69)s and the life time(2 455.0±263.5)s of the sperm both reached the maximum values.Sperm of L.waleckii could be activated by NaCl solutions within the concentration range of 0.017 1-0.085 5 mol·L-1.The longest duration of fast movement(294.33±21.42 s)was observed in 0.085 5 mol·L-1 NaCl group,while the maximum life time(933.33±77.11 s)occurred in 0.056 4 mol·L-1 NaCl group.Activation by KCl solutions resulted in relatively shorter durations for both fast movement and life time.The peak duration of fast movement(231.67±29.76 s)was achieved in 0.1 mol·L-1 KCl group,and the maximum life time(367.0±12.53)s was recorded in 0.05 mol·-1 KCl group.Sperm motility was poor in MgCl2 solutions,with the highest life time reaching only 100 s.When activated by 100 mmol·L-1 NaHCO3,sperm exhibited the longest duration of fast movement(657.0±20.53 s),and the life time(2 414.00±87.47 s)was achieved in 75 mmol·L-1 NaHCO3 group.Glucose solution at 0.1 mol·L-1 also yielded favorable sperm motility.The study demonstrates that Dali Lake water,pH 8,NaCl(0.056 4-0.085 5 mol·L-1),NaHCO3(75-100 mmol·L-1),glucose(0.1 mol·L-1),and KCl(0.1 mol·L-1)can each function as a sperm motility activating agent for L.waleckii.However,sperm activated by Dali Lake water or 100 mmol·L-1 NaHCO3 exhibited superior motility,whereas activation with KCl and MgCl2 solutions results in comparatively poor motility.These findings can provide valuable reference for selecting activating agents during the artificial propagation of L.waleckii in Dali Lake.关键词
