基于线粒体12S rRNA的南极鱼亚目鱼类物种鉴定研究
Identification analysis of DNA barcoding based on mitochondrial 12S rRNA gene in Notothenioidei fishes

Abstract
Notothenioidei fishes are diverse and the morphological differences between families are not obvious.In order to facilitate the exploration of species diversity among Notothenioidei,and make accurate species identification,101 bycatch Notothenioidei fishes from Antarctic krill fishery were collected in CCLAMR subareas 48.1 and 48.2 from 2021 to 2023.We analyzed the phylogenetic relationships among Notothenioidei fishes,and verified the applicability of 12S rRNA gene as a DNA barcode for the species identification of Notothenioidei fishes.Partial 12S rRNA sequences of 101 Notothenioidei fishes were obtained by PCR amplification and sequencing.These amplified sequences were subsequently used for phylogenetic analyses alongside sequences from 13 species in 13 genera from the GenBank database.To analyze the evolutionary relationships among 13 Notothenioidei fishes,we constructed the NJ tree and mapped haplotype networks.The results indicated that the 101 samples belonged to 13 species,13 genera,and 3 families.The average inter-species genetic distances based on 12S rRNA was 0.0391,which was 11.7 times the average intra-species genetic distance,and met the basic requirements for DNA barcode.Phylogenetic analysis indicated that except for Chaenocephalus aceratus and Chionodraco rastrospinosus,the remaining species could form independent branches and species of the same family clustered into one branch.In summary,12S rRNA gene can be used as an effective DNA barcode for identifying Notothenioidei species,and shows clear evolutionary relationships at family level.关键词
