Molecular identification of parasites in a Neophocaena asiaeorientalis sunameri stranded at the Yangtze Estuary

Abstract

The East Asian finless porpoise(Neophocaena asiaeorientalis sunameri)is a national second-class protected wild animal,and the Yangtze River Estuary is home to a population of Neophocaena asiaeorientalis sunameri.During the necropsy of an East Asian finless porpoise stranded at the Yangtze River Estuary,three parasite nodules(2-3 cm in diameter)were found in the muscles surrounding the genital slit.Three parasites from each nodule were separated carefully,each was measured to be approximately 10 cm in length.To identify the species of parasites,molecular identification was conducted on three parasitic samples.After morphological observation,the genomic DNA of the isolated parasites was extracted and internal transcribed spacer(ITS)gene fragments were amplified by PCR.The PCR results showed that the amplification products of ITS gene were 1 500 bp approximately.Blast alignment results showed that the identities with reported nematodes in the NCBI Genebank reached up to 99%-100%.Phylogenetic analyses based on the ITS2 gene showed that the parasite samples isolated in this study were genetically closest to Crassicauda fuelleborni with a bootstrap value of 96%,and they were on the same branch.In summary,the isolated parasite samples should be Crassicauda fuelleborni,belonging to Spirurina and Tetrameridae.The results can enrich the genotyping data of Crassicauda from Neophocaena asiaeorientalis sunameri and provide reference for future research.
