多裂叶水芹线粒体特征及雄性不育候选基因的鉴定
Identification of mitochondrial characteristics and candidate male sterile genes in Oenanthe thomsonii

Abstract
[Objectives]The paper aimed to analyze the mitochondrial characteristics of Oenanthe thomsonii and identify the candidate genes for male sterility,providing a reference for the hybrid breeding and theoretical research of male sterility in water dropwort.[Methods]With the male sterile of O.thomsonii(named OtBY002)and corresponding fertile lines(named OtKY002)as materials,the mitochondrial genomes were sequenced and assembled using the second-generation sequencing platform provided by Ⅲumina and the third-generation sequencing platform provided by Nanopore.Then,the genomes were annotated,candidate genes were screened and obtained,and three-dimensional structure analysis and evolutionary analysis of the protein of the sterility gene were conducted.[Results]OtBY002 and OtKY002 both contained one large and one small loop mitochondrial genome,all containing 75 genes.Among them,the large mitochondria genome lengths of OtBY002 and OtKY002 were 336 668 bp(accession number:PP693295.1)and 336 731 bp(PP693297.1),respectively,the two small mitochondria genome sizes were the same and both 48 051 bp in length(accession number:PP693296.1 and PP693298.1,respectively).Further analysis indicated that the length of the ccmFN gene sequence showed significant differences between OtBY002 and OtKY002,with 1 050 and 1 740 bp,respectively.The mutation of the OtBY002 gene at 921 bp was the causing infertility,with a certain evolutionary distance between the ccmFN of OtBY002 and OtKY002,which was not located on the same branch.[Conclusions]The ccmFN gene sequence was the cause of OtBY002 male sterile.关键词
