联合转录组和代谢组分析光照对葡萄果实成熟的影响
Combined transcriptome and metabolome analysis of the effects of light on grape fruit ripening

Abstract
[Objectives]The paper aimed to understand how light affects grape fruit growth and maturation through transcriptional regulation.[Methods]Using'Kyoho'grape fruit as the experimental material,the fruits were covered with black bags for shading at 100 d after flowering,while others were exposed to normal light as controls.The physiological and quality indicators of the fruits related to maturity were measured.Transcriptome and metabolome sequencing were performed on the skin and flesh of shaded and normally-light fruit,in each of the two comparison groups:shaded skin vs light-treated skin and shaded flesh vs light-treated flesh,focusing on gene expression differences in flavonoid,aroma metabolic and chlorophyll synthesis pathways.[Results]Shading treatment increased the contents of chlorophyll,titratable acids,starch,and sucrose synthase activity in fruits but decreased the contents of anthocyanins,glucose,fructose,and the activities of sucrose phosphate synthase,acid invertase,and neutral invertase compared to light treatment.These differentially expressed genes were mainly involved in plant hormone synthesis,starch and sucrose metabolism,and secondary metabolite synthesis.Pathways related to fruit color formation,such as carotenoid and flavonoid biosynthesis,were also significantly enriched.[Conclusions]Shading treatment reduced anthocyanin contents and increased chlorophyll contents,inhibiting normal fruit coloring.Light treatment regulated key genes in pigment and sugar metabolism pathways,significantly affecting fruit quality.关键词
