Abstract

[Objectives]Meloidogyne enterolobii spreads quickly,which has a wide host range,and is highly pathogenic,causing substantial economic losses annually.To effectively control its damage,highly effective Bacillus strains for controlling Meloidogyne enterolobii were screened in vitro and their practical application effect were verified.[Methods]In vitro nematicidal assays were employed to screen strains with nematicidal activity.Meanwhile,the biocontrol strains were classified and identified by combining morphological,physiological,biochemical characteristics and molecular biological methods.And the activities of serine protease and metalloprotease of them were detected.Subsequently,their control effects were evaluated through pot experiments.[Results]Six Bacillus strains with good effects were screened,namely strains 3313-1,1021-4,752-3,2968-1,755-2,1439-1.Based on the morphological characteristics,physiological and biochemical characteristics and molecular biological identification results,strains 3313-1,2968-1 and 1021-4 were B.mycoides,strain 1439-1 was B.halotolerans,strain 755-2 was B.cereus,and strain 752-3 was B.thuringiensis.The adjusted mortality rates of nematodes treated with their fermentation broths were all greater than 80%,and the adjusted mortality rates of nematodes treated with their fermentation supernatants were all above 60%.Moreover,all six strains possess the activity of hydrolyzing proteins.Among them,strains 3313-1,1021-4 and 752-3 were also capable of producing serine protease and metalloprotease.The results of pot experiment indicated that the galling indexes of the six strains were significantly lower than that of the blank control(P<0.05)and showed little different from that of the positive control(41.7%fluopyram suspension concentrate).The control effect of each strain was more than 50%,and the control effect of strains 1439-1 and 752-3 were better than that of the positive control,with control effects of 71.70%and 72.45%,respectively.[Conclusions]In this experiment,169 Bacillus strains were successfully isolated from 9 soil samples.Through contact-killing tests on J2 using the fermentation broth and fermentation supernatant of strains,six Bacillus strains with relatively good control effects in both in vitro nematicidal tests and practical applications were finally screened out.This will lay a foundation for the subsequent isolation of nematoidal active ingredients,the research on nematoidal mechanism,and the development of microbial nematode-killing products.
