Abstract

[Objectives]Polysaccharide is one of the main active components of the medicinal plant Morinda officinalis,and its synthesis is closely related to the sucrose synthase(SUS)gene family.In this study,members of the SUS gene family of M.officinalis were identified and their expression patterns in roots of different years were analyzed,providing the theoretical basis for mining the SUS genes family related to polysaccharide synthesis of M.officinalis.[Methods]Based on the transcriptome data of M.officinalis,the SUS gene family members were predicted and mined by sequence comparison and hidden Markov model,and the physicochemical properties and structural characteristics of these genes were analyzed by TBtools and other bioinformatics tools.MoSUS genes derived from transcriptome data were combined with six AtSUS genes of Arabidopsis thaliana using MEGA 11 to construct a phylogenetic tree.[Results]The length of amino acids encoded by 18 MoSUS ranged from 192 to 902 amino acids and their relative molecular weight ranged from 21.53×103 to 102.14×103,which were theoretically acidic proteins.Subcellular localization predicted that most MoSUS proteins were localized in the cytoplasm.MoSUS was divided into three subfamilies according to the phylogenetic tree,with SUS Ⅰ and SUS Ⅱ subfamilies each possessing eight members respectively and SUS Ⅲ subfamily possessing two members.The members of the SUS Ⅰ and SUSⅢ subfamilies showed high sequence conservation.MoSUS1 and MoSUS2,MoSUS5 and MoSUS6 contained the same number and order of motifs.Gene expression analysis showed that expression of MoSUS13-MoSUS1 7 genes increased with the age of the samples.[Conclusions]The phylogenetic analysis showed that the SUS genes in M.officinalis were divided into three subfamilies:SUS Ⅰ,SUS Ⅱ and SUSⅢ.MoSUS13-MoSUS17 might be involved in regulating the synthesis of polysaccharides.The results of this study laid a foundation for further study on the function of SUS genes of M.officinalis,and also provided a scientific basis for research on the formation mechanism of the medicinal quality of M.officinalis.
