基于茎尖脱毒的宁夏枸杞无病毒种苗制备技术研究
A Study on Preparation Technology of Ningxia Lycium barbarum Virus-Free Seedlings Based on Shoot Tip Detoxification

Abstract
[Objective]To address the issue of germplasm degradation of Ningxia Lycium barbarum caused by viral diseases,and to establish an efficient stem tip virus-free technology system,providing technological support for the large-scale production of virus-free seedlings.[Methods]Using the stem tips of"Ningqi No.1"as explants,and taking survival rate,germination rate,redifferentiation rate and detoxification rate as evaluation indicators,the hormone ratio of 6-BA(0.05-0.5 mg·L-1)and NAA(0.01-0.2 mg·L-1)in MS medium was systematically optimized to screen the optimal stem tip length.And the detoxification effects of each treatment were detected by ELISA.[Result]When the medium was a combination of 0.2 mg·L-1 6-BA+0.05 mg·L-1 NAA,the induction rate of tissue culture seedlings was the highest,reaching 73.33%±6.7%;When the concentration of NAA was 0.1 mg·L-1,the differentiation rate reached 80%±6.7%,and the root development was optimal(P<0.05).The length of the stem tip was significantly negatively correlated with the detoxification efficiency(r²=0.904 3).Stem tips with a length of 0.21-0.30 mm had both a high survival rate(62.2%±3.8%)and a high detoxification effect(82.2%±3.8%).[Conclusion]By optimizing the hormone ratio and the length of the stem tip,The virus-free seedling preparation technology established for Ningxia Lycium barbarum has a virus removal rate of over 82%and a transplanting survival rate of over 65%,providing a standardized solution for the prevention and control of Lycium barbarum viruses and the improvement of their quality.关键词
