Abstract

Objective To investigate the effects of Honey-processed Radix Hedysari+vinegar-processed Rhizoma Curcumae(炙红芪-醋莪术,RHRC)on intestinal immunity in mice with colitis-associated colorectal cancer(CAC).Methods 40 C57BL/6 mice were randomly divided into the control group,azoxymethane/dextran sodium sulfate group(model group),sulfasalazine positive control group,and the RHRC(4∶1)group.The CAC model was induced by AOM and DSS in the other groups.The rats in the positive control group and the drug group were treated with sulfasalazine suspension and RHRC(4∶1)extract,respectively,from the beginning of the experi-ment.DAI scores were calculated,and spleens were removed for determination of the expression of T cell subsets CD4+,CD8+.The CD4+/CD8+ratio in the spleen of the model group was significantly decreased.Results Compared with the blank group,the percentage of CD4+T cells and CD4+/CD8+ratio in the spleen of the model group were significantly decreased(P<0.01),and the percentage of CD19 B cells was significantly decreased(P<0.01).Compared with the model group,the percentage of CD4+T cells,CD4+/CD8+ratio and CD19 B cells in the spleen of the HRCR-treated group were significantly increased(P<0.05 or 0.01).Conclusion RHRC can increase the ratio of CD4+/CD8+and the percentage of CD19 B cells,ameliorate the immune deficiency caused by CAC,and main-tain the immune balance.The mechanism may be related to SCFAs in activating the GPR43-mediated ERK-NF-κB signaling path-way.
