淫羊藿苷对LPS诱导的滑膜成纤维细胞炎症反应、自噬及纤维化的调控作用
Regulatory effects of icariin on the LPS-induced inflammatory response,autophagy,and fi-brosis in synovial fibroblasts

Abstract
Objective To investigate the regulatory effect of icariin on lipopolysaccharide(LPS)-induced inflammatory response,auto-phagy activity,and fibrosis in rat synovial fibroblasts(SFs).Methods SFs were treated with different concentrations of icariin,and cell proliferation activity was determined by MTT assay to determine optimal action concentration.Cells were divided into the control group,model group(10 μg/mL LPS),icariin group(20 μmol/L),YY-20394 group(100 μmol/L),and icariin+YY-20394 group(20μmol/L+100 μmol/L).The contents of IL-1β,TNF-α and IL-6 inflammatory factors were determined by ELISA,while the ex-pressions of autophagy-related proteins Beclin-1 and ATG7,fibrosis marker proteins COL1A1 and TIMP1,and PI3K,p-AKT and p-NF-κB signaling pathway proteins were determined by Western blot(WB).Results Compared with the control group,the model group exhibited significantly concentrations of inflammatory factors of IL-1 β,TNF-α,and IL-6 in SFs,along with increased expres-sion of autophagy-related proteins Beclin-1 and ATG7,fibrosis marker proteins COL1A1 and TIMP1,PI3K,p-AKT and p-NF-κB signal pathway protein(P<0.05).Compared with the model group,both the icariin and YY-20394 groups showed reduced con-centrations of the above inflammatory factors and protein expressions of autophagy-,fibrosis-,and pathway-related proteins(P<0.05).Further comparison was made between the icaritin and YY-20394 single treatment groups,the SFs in icariin+YY-20394 group not only decreased the concentrations of inflammatory factors of IL-1 β,TNF-α and IL-6,but also significantly decreased the expressions of Beclin-1,ATG7,COL1A1,TIMP1,PI3K,p-AKT and p-NF-κB proteins(P<0.05).Conclusion Icariin signifi-cantly inhibits LPS-induced inflammatory response,autophagy activity,and fibrosis in rat SFs,potentially mediated through the key PI3K/AKT/NF-κB signaling pathway.关键词
