四物五子丸含药血清对过氧化氢诱导的人视网膜色素上皮细胞氧化损伤的保护作用
Protective effect of Siwu Wuzi Pill(四物五子丸)containing serum on hydrogen peroxide-in-duced oxidative damage in ARPE-19 cells

Abstract
Objective To observe the protective effect of Siwu Wuzi Pill(四物五子丸,SWWZP)containing serum on hydrogen perox-ide(H2O2)-induced oxidative damage in human retinal pigment epithelial(ARPE-19)cells.Methods H2O2 induced the oxidative damage in ARPE-19 cells,which were then assigned to the blank control group,model control group,and SWWZP-containing serum intervention group(further divided into low,medium,and high dose groups);MTT method,scratch test and colorimetric method were used to detect MDA and CAT;Western blot(WB)was used to detect the expressions of Cat B,Cys C,MMP-2 and TIMP-2 proteins;and PCR was used to detect the expressions of Cat B,Cys C,MMP-2 and TIMP-2 mRNA.Results Compared with the blank control group,H2O2 at a concentration of 200 μmol/L formed oxidative damage to ARPE-19 cells for 24 h(P<0.05);SWWZP-containing serum at concentrations of 2.5%,5%and 10%had a proliferation-promoting effect on ARPE-19 cells(P<0.05),and the serum intervention group increased the viability of ARPE-19 cells(P<0.05),decreased the MDA content(P<0.001),increased the CAT activity(P<0.05),and significantly decreased the high expression levels of Cat B,Cys C,MMP-2,and TIMP-2(P<0.05),and significantly down-regulated the high expression levels of Cat B,Cys C,MMP-2,and TIMP-2 mRNA(P<0.01).Conclusion SWWZP-containing serum was able to increase the antioxidant capacity of ARPE-19 cells,thus exerting a protective effect on ARPE-19 cells.关键词
