参附注射液调控Nrf2信号通路介导的中性粒细胞诱捕网途径对脓毒血症肾损伤的保护作用研究
Protective effect of Shenfu Injection(参附注射液)on sepsis-associated acute kidney injury through regulating the Nrf2 signaling pathway mediated by neutrophil extracellular traps(NETs)formation

Abstract
Objective To explore the protective effect of Shenfu Injection(参附注射液,SFI)on sepsis-associated acute kidney injury(AKI)and its mechanism through regulating the Nrf2 signaling pathway to control the formation of neutrophil extracellular traps(NETs).Methods 30 C57BL/6 mice were randomly divided into six groups:the control group(CON),sepsis group(CLP),Nrf2 in-hibitor group(siNrf2),normal saline group(NS),SFI treatment group(SA),and SFI+Nrf2 inhibitor group(SA+siNrf2).Sepsis was induced by cecal ligation and puncture(CLP),and the body weight,renal function,inflammatory factors,and NETs levels of the mice in each group were observed.Results Mice in the CLP group exhibited typical sepsis symptoms,including weight loss,dispirited-ness,decreased appetite,and dry fur.Mice in the siNrf2 group showed further exacerbation of these symptoms,while the SA group showed significant alleviation.Compared with the CLP group,the siNrf2 group exhibited worse renal function,with significantly reduced Nrf2-related protein expression and significantly increased inflammatory factors and NETs markers(P<0.05).The SA group showed amelioration in these indicators(P<0.05).Compared with the NS group,the SA+siNrf2 group showed significantly improved renal function and reduced inflammatory status,increased Nrf2-related protein expression,and reduced NETs formation(P<0.05),but the protective effect was weaker than that of the SA group(P<0.05).Conclusion SFI exerts a protective effect on sepsis-associated AKI by activating the Nrf2 signaling pathway,inhibiting NETs formation,improving the renal function,and reducing inflammation.关键词
