Abstract
To investigate the effects of several organic nitrogen sources on the fermentation potency of avermectin and explore methods for reducing the production cost of avermectin.Parallel experiments in 50 L fermenters were conducted to prove their effects on potency by replacing soybean cake powder and yeast powder with peanut meal,replacing soybean cake powder with soybean meal powder,replacing starch with com meal,and replacing soybean cake powder with animal peptone.The results showed that:peanut meal at concentrations of 12.8 g·L-1 and 5.8 g·L-1was used to replace soybean cake powder and yeast powder,the average final titers were reduced by 40%and 22%compared to the control,with significant reductions(P=6.1×10-5).Soybean meal powder at 23 g·L-1 was used to completely replace soybean cake powder,resulting in an average final potency increased by 9%compared to the control group.However,after adding 6%soybean oil,the average fermentation potency was 32 μg·g-1 lower than the control group.There was no significant difference between the two group(P=0.92).Starch and soybean cake powder were replaced with corn meal,at concentrations of corn meal(60 g·L-1),starch(98 g·L-1)and soybean cake powder(17 g·L-1 or 20 g·L-1),resulting in an average final potency 12%lower than the control group,showing a significant decrease(P=0.016).Animal peptone at 4 g·L-1 was used as an equivalent substitute for soybean cake powder,resulting in a final potency 21%lower than the control group.In conclusion,soybean meal powder,when used as a replacement for soybean cake powder,can increase the avermectin fermentation potency and reduce production cost.Peanut meal adversely affects avermectin biosynthesis,when its negative impact becoming increasingly detrimental at higher concentrations,therefore,it is not suitable for use in the fermentation medium.The strategy of replacing starch with corn meal to bypass starch processing and lower costs is not viable.Animal peptone disrupts mycelial aggregation,thereby reducing fermentation potency.It is not recommended as a substitute for soybean cake powder in avermectin fermentation.关键词
