青砖茶水提物对小鼠急性胃溃疡保护作用研究
Study on the Protective Effect of Qingzhuan Dark Tea Water Extract on Acute Gastric Ulcers in Mice

Abstract
Objective To investigate the protective effect of Qingzhuan Dark Tea Water Extract(QDTE)on acute gastric ulcer in-duced by absolute ethanol(AE)in mice.Methods Forty male Kunming mice were randomly divided into control group,model group,posi-tive control group(50 mg/kg sucralfate,sucralfate group),and QDTE administration group(2.56 g/kg QDTE,QDTE group).Control group and model group were given normal saline,while the other two groups were given the corresponding drug doses once a day for 7 consecutive days.Two hours after the last administration,except for the control group which received an equal volume of normal saline,the other three groups were orally administered AE(10mL/kg)to establish an acute gastric ulcer model.One hour later,the samples were collected,and the ulcer index and ulcer inhibition rate were calculated.The contents of TNF-α,IL-6,COX-2,and PGE2 in serum were determined.The morphology and pathological changes of gastric tissue were observed by HE and PAS staining.The expression level of epidermal growth factor(EGF)protein in gastric tissue of each group was measured by immunohistochemistry.Results Compared with the control group,the ulcer index of mice in the model group significantly increased,the ulcer inhibition rate was 0,and the contents of TNF-α,IL-6,COX-2,and PGE2 in serum increased(P＜0.01).HE staining showed that the glandular structure of gastric mucosa in model group was severely damaged,ac-companied by a large number of inflammatory cell infiltrations.PAS staining showed that the positive substances disappeared and the mucin was severely damaged.Compared with the model group,QDTE intervention reduced the ulcer index,increased the ulcer inhibition rate,sig-nificantly reduced the contents of TNF-α,IL-6,COX-2,and PGE2 in serum,and increased the positive cell expression of EGF in gastric mu-cosa tissue(P＜0.05).Conclusion QDTE has a certain preventive and protective effect on acute gastric ulcer in mice,which may be relat-ed to reduce the expression levels of inflammatory factors and promote of gastric mucosal regeneration and repair.关键词
