摘要
Abstract
Objective To evaluate the inhibitory effect and partial molecular mechanism of betulinic acid(BA)on the malignant bi-ological behavior of cervical cancer SIH A cells.Methods Cervical cancer SIH A cells were cultured in cell incubator with constant tempera-ture and humidit and treated with different concentration gradients of BA to detecte the proliferation and migration ability.The optical-density of SIHA cells was detected by MTT assay,and then the time-dependent inhibition of SIHA cells by BA at the same concentration was evalua-ted.The single cell clonal proliferation ability of SIHA cells was detected by plate clone formation assay.The migration and invasion ability of SIHA cells were evaluated by Transwell cell small chamber migration and invasion assay.The c-Myc protein expression level in SIHA cells treated with BA was deteced by Western blot assay.Resue assay was applied to demonstrate whether the inhibitory effect of BA was mediated by c-Myc pathway.Results BA could inhibit the survival and proliferation of SIHA cells in a time and dose-dependent manner,as well as the migration and invasion in a dose-dependent manner.In a certain dose range,BA down-regulated the expression of oncogene c-Myc protein.c-Myc-saRNA can partly counteract the inhibition effect of BA.Conclusion BA can inhibit the proliferation and migration of cervical cancer SIHA cells,and its mechanism is related to the down-regulation of c-Myc protein expression.关键词
