Abstract

Objective To use network pharmacology,molecular docking technology,and cell experiments to deeply explore the mech-anism of trifluoperazine dihydrochloride on prostate cancer.Methods The targets of trifluoperazine dihydrochloride and prostate cancer-relat-ed targets were screened out through the database and a visual network and protein-protein interaction network(PPI)were constructed,fol-lowed by GO and KEGG enrichment analysis was carried out,and finally,the potential targets were verified by molecular docking software.In the cell experiment,prostate cancer cells were treated with trifluoperazine dihydrochloride at different concentration gradients.The MTT col-orimetric method and plate clone formation experiment were used to verify its inhibitory effect on cell proliferation.The scratch experiment and transwell invasion and migration experiment were employed to demonstrate the intervention effect on the invasion and migration charac-teristics of cancer cells.Results The results of network pharmacology analysis showed that trifluoperazine dihydrochloride has 70 targets in-volved in the fight against prostate cancer.KEGG enrichment analysis further revealed that the drug can exert its significant anti-prostate cancer effect through multiple key signal pathways such as cancer-related pathways and PI3K-AKT pathway.The molecular docking results showed that trifluoperazine dihydrochloride has an extremely ideal binding activity with the core targets.And its inhibitory effect on the vitali-ty,proliferation,migration and invasion of prostate cancer cells showed a distinct dose-dependent characteristic.Conclusion Trifluoperazine dihydrochloride may inhibit prostate cancer cell proliferation through multiple targets,and the inhibitory ability is proportional to the dose.
