Abstract
Objective To observe the expression of tumor protein 53(P53),cytokeratin 5/6(CK5/6),and human epidermal growth factor receptor 2(HER-2)in breast cancer tissues and analyze their associations with pathological features,as well as to evaluate survival curves based on different expression levels.Methods A total of 120 breast cancer patients who underwent radical resection in Gongyi People's Hospital from January 2021 to September 2023 were selected.Immunohistochemistry was used to detect the expression of P53,CK5/6,and HER-2 in breast cancer tissues and adjacent noncancerous tissues.The associations of P53,CK5/6,and HER-2 expression with clinicopathological characteristics were analyzed.Kaplan-Meier curves were used to calculate survival curves according to the expression status of P53,CK5/6,and HER-2.Results The positive expression rates of P53,CK5/6,and HER-2 were higher in breast cancer tissues than in adjacent tissues(P<0.05).Compared with P53-negative patients,P53-positive patients showed statistically significant differences in age,tumor size,lymph node metastasis,and vascular tumor thrombus(P<0.05),while there were no significant differences in TNM stage,perineural invasion,or vascular invasion(P>0.05).Compared with CK5/6-negative patients,CK5/6-positive patients had significant differences in histological grade and lymph node metastasis(P<0.05),but not in age,tumor size,TNM stage,perineural invasion,vascular invasion,or vascular tumor thrombus(P>0.05).Compared with HER-2-negative patients,HER-2-positive patients had significant differences in age,histological grade,lymph node metastasis,vascular invasion,and vascular tumor thrombus(P<0.05),but not in tumor size,TNM stage,or perineural invasion(P>0.05).Spearman correlation analysis showed that positive P53 expression was positively correlated with lymph node metastasis(r=0.268,P=0.003)and vascular tumor thrombus(r=0.225,P=0.014).Positive CK5/6 expression was positively correlated with high-grade histological differentiation(poorly differentiated)(r=0.429,P<0.001),lymph node metastasis(r=0.265,P=0.003),vascular invasion(r=0.272,P=0.003),and vascular tumor thrombus(r=0.218,P=0.016).Kaplan-Meier curves showed that patients with positive expression of P53,CK5/6,or HER-2 had shorter overall survival compared with negative patients(P<0.05).Conclusion The expression of P53,CK5/6,and HER-2 is associated with invasive features and poor prognosis of breast cancer.Combined detection may have clinical reference value,but this conclusion requires confirmation through studies with larger sample sizes and more rigorous designs.关键词
