摘要

Abstract

This study aimed to evaluate the immune duration of a biomineralized foot-and-mouth disease virus-like particle(FMDV VLPs)vaccine.Using purified O-type FMDV VLPs antigen as the founda-tion,a calcium phosphate biomineralized protective layer was constructed on the particle surface through biomineralization.Vaccines were formulated by combining the biomineralized VLPs with ISA 201 adjuvant or water-soluble adjuvant,followed by immunization of swine.Lymphocyte proliferation,Th1/Th2 immune responses,specific antibody titers,and protection efficacy post-challenge were as-sessed to evaluate the vaccine's immune duration.Results showed that at 28 days post-immunization,both biomineralized FMDV VLPs formulated with ISA 201 adjuvant or water-soluble adjuvant induced Th1/Th2 immune responses,lymphocyte proliferation,and high-titer specific antibody production.By 210 days post-immunization,the ISA 201 adjuvant group maintained specific antibody titers above 1∶64,achieving an 80%protection rate post-challenge.This study demonstrates that biomineralized FMDV VLPs combined with ISA 201 adjuvant can provide long-lasting immune protection for at least 210 days,offering a novel strategy for developing safe and stable FMDV VLPs vaccines.
