摘要

Abstract

This study investigated the immunogenicity of Salmonella outer membrane protein PgtE.The pgte gene was cloned into pET28a,then PgtE proteins were expressed in Escherichia coli BL21,and were purified via His-tag affinity chromatography.PgtE proteins were mixed with manganese gluconate(Mn2+)and aluminum hydroxide(AH)adjuvant to immunize mice at the dosage of 50 μg per mouse.Cell proli-feration assays,enzyme-linked immunosorbent assay(ELISA),and bacterial load tests were used to evaluate the immunological effects.Compared with groups,the data showed that PgtE+Mn2++AH group signifi-cantly enhanced splenic lymphocyte proliferation(SI),elevated levels of IL-2,IL-4,IL-10,IFN-γ and IgG(P<0.05)in serum,and reduced Salmonella loads in the spleen,liver,and kidney(P<0.05).PgtE+Mn2++AH group and PgtE group exhibited the immunoprotection rate of 100%and 77%,respectively,which was significantly higher than PBS group.These results indicate that PgtE exhibits strong immunogenicity,Mn2+and AH adjuvant synergistically enhances its immune efficacy,providing a foundation for Salmonella subunit vaccine development.
