Abstract

In the current toxicity studies of triclocarban,reproductive toxicity has attracted much attention.In order to explore the reproductive toxicity of triclocarban,rats were selected as experimental animals to study the possible damage of triclocarban to testicular tissue in male rats.Five-week-old SD rats were used as experimental animals,and triclocarban weight:diet weight ratio of 1∶1 000,2∶1 000,8∶1 000 groups and control group were set up,and the feeding period was 45 days.Enzyme-linked immunosorbent assay(ELISA)was used to detect the contents of testosterone,estradiol,luteinizing hormone and follicle-stimulating hormone in serum.Immunohistochemistry assay was used to determine the expression levels of Bax and Bcl-2 in testis.The expression levels of FAS,FASL,FADD,Bcl-2,Bax,CREM and P53 genes in rat testicular tissues were detected by fluorescence quantitative PCR.Western-blot was used to detect the related apoptotic proteins.The results showed that compared with the control group,the rats exposed to triclocarban decreased body weight,increased the coefficient of sexual organs,endocrine disorders of sex hormones,decreased sperm quality and quantity,and pathological changes such as testicular atrophy and edema.Pathological sections showed epithelial shedding,vacuolization and decreased sperm motility in the seminiferous tubules.The TUNEL assay showed that the fluorescence sites of apoptotic cells appeared in spermatogonia,spermatogenic cells at all levels,and supporting cell locations.There were no significant changes in the serum levels of testosterone in the 2∶1 000 and 8∶1 000 groups,estradiol levels in the 8∶1 000 group,and the hormone levels of follicle-stimulating hormone decreased.The mRNA expression level of FADD in the 2∶1 000 and 8∶1 000 groups was significantly increased.The gray values of protein bands of FADD,Caspase-8 and Caspase-3 increased with increasing doses of triclocarban exposure.The changes in the protein levels of Caspase-9,Cytochrome C and NF-κB p53 were similar to those in the testes and epididymis of rats.Triclocarban can cause damage to the testes of rats,including inflammation,hormonal disorders and testicular germ cell apoptosis,which may be due to the fact that triclocarban can regulate testicular cell apoptosis through FADD-related signaling pathways.
